What does GHRP-6 do?
GHRP-6 (Growth Hormone Releasing Peptide-6) is a synthetic hexapeptide that prompts the release of growth hormone in research. It does so not by being growth hormone itself, but by sending a signal: it binds to the GHS receptor, the same receptor on which the hunger hormone ghrelin acts, and thereby prompts the pituitary to release growth hormone. GHRP-6 is one of the first GHRPs developed and is often used as a reference substance in the literature.
How does GHRP-6 work?
GHRP-6 is a ghrelin mimetic: it mimics the action of ghrelin, the hormone your body produces when you are hungry. By binding to the GHS receptor (growth hormone secretagogue receptor) in the pituitary and hypothalamus, it sends a pulse that prompts the pituitary to release growth hormone. In research, GHRP-6 is therefore called a secretagogue, a substance that boosts the body-own release rather than supplying the hormone itself. What we know about this comes from the scientific literature.
GHRP-6 and appetite
The most striking feature of GHRP-6 is its effect on appetite. Because it acts on the same receptor as ghrelin, the hunger hormone, it is associated in research with a clear increase in hunger. That sets GHRP-6 apart from many later peptides in the same family: where ipamorelin, for example, is selective and barely affects appetite, GHRP-6 is known as the "hunger variant". GHRP-6 vs. GHRP-2 and ipamorelin
Within the family of growth hormone secretagogues, GHRP-6 is often compared with GHRP-2 and ipamorelin. The difference is mainly in two things: appetite and selectivity. GHRP-2 gives a strong growth-hormone response with less hunger than GHRP-6. Ipamorelin goes one step further and is the most selective of the three: it stimulates growth-hormone release without notably affecting appetite, cortisol or prolactin. GHRP-6 is therefore the oldest and least selective, but precisely for that reason of interest in research around the ghrelin route. Side effects of GHRP-6
In the research literature, GHRP-6 is described with a few typical effects related to its action on the ghrelin and growth-hormone route. The most commonly mentioned are a strong sense of hunger, fluid retention (oedema) and a temporary rise in cortisol and prolactin, the latter is exactly where it differs from the more selective ipamorelin. Sometimes a feeling of warmth, mild nausea or flushing are also mentioned. These are observations from the literature, not data about laboratory use. Reconstituting and storing GHRP-6
GHRP-6 is supplied as a lyophilised powder in a vial. For research it is mixed with bacteriostatic water; GradePurity gives no dosing or administration advice. After mixing, the solution goes in the fridge (2–8 °C), protected from light, and is used within a short time; for longer storage the unopened vial can go in the freezer.
