What does retatrutide do?
Retatrutide is the metabolic research peptide everyone is talking about right now, mostly because of the striking weight-loss results in Eli Lilly's large Phase 3 research. It is a so-called triple agonist: it works on three "switches" in the body at once, GLP-1, GIP and glucagon, which control hunger, blood sugar and energy expenditure. It is thought that this very threefold action takes retatrutide a step beyond older options such as semaglutide (Ozempic) and tirzepatide.
How does retatrutide work?
Put simply, retatrutide works on three pathways at once. The GLP-1 and GIP pathways curb appetite and help keep blood sugar balanced; the glucagon pathway gives energy expenditure an extra push on top. No other known weight-loss peptide has that third pathway, and it is thought the combination therefore goes further than single GLP-1 options. The full action is being studied in Eli Lilly's Phase 3 TRIUMPH programme; as a medicine retatrutide is not yet approved.
Retatrutide vs. Ozempic
The difference in one sentence: Ozempic (semaglutide) presses one switch, retatrutide presses three. Ozempic is a single GLP-1 agonist; retatrutide also activates GIP and glucagon. Tirzepatide (Mounjaro) sits in between with two pathways. So retatrutide is not the same as Ozempic, but its own, newer research peptide, studied precisely for that extra mechanism.
Side effects of retatrutide
In the Phase 3 research, the most commonly reported reactions were gastrointestinal, nausea, reduced appetite, sometimes diarrhoea, usually mild and mostly early on, much like other GLP-1 options.
